چکیده

The enzymatic reduction of /Shydroxy-P-methylglutaryl coenzyme A to mevalonic acid has been measured in cellfree systems prepared from the livers of rats maintained under various dietary conditions. It has been shown that the reductase reaction is markedly depressed when cholesterol is fed. This diminished activity has been observed in both the microsomes and in the subsequently solubilized preparation when an acetone powder technique is used. The effect of fasting upon fl-hydroxy-/3-methylglutaryl coenzyme A reduction has been examined and the results also show a dramatic decrease in the reductase activity in both the microsomes and the solubilized enzyme.

Effect of pravastatin on biliary lipid composition and bile acid synthesis in familial hypercholesterolaemia.

Nine patients with heterozygous familial hypercholesterolaemia were treated for eight weeks with either 40 mg pravastatin or placebo under double blind conditions. Six patients received pravastatin, a competitive inhibitor of 3-hydroxy-3-methylglutaryl-coenzyme A (HMG-CoA) reductase. Treatment with pravastatin resulted in a significant decrease in plasma cholesterol caused by a decrease in low ...

Effect of Standardized Amla Extract on Atherosclerosis and Dyslipidemia

Emblica officinalis, commonly known as Indian gooseberry (Amla), is found to be effective for the reversal of dyslipidemia and intima-media thickening and plaque formation in the aorta in hypercholesterolaemic rabbits. In this study, cholesterol powder (100 mg/kg body weight) was administered orally to healthy NZ white rabbits for 4 mo to induce hypercholesterolaemia; and thereafter, amla extra...

Properties of 3-hydroxy-3-methylglutaryl coenzyme A reductase solubilized from rat liver and hepatoma.

In hepatomas, the activity of 3-hydroxy-3-methylglutaryl coenzyme A reductase, the rate-controlling enzyme in cholesterol biosynthesis, is not normally suppressed by cholesterol. To examine the biochemical mechanism of this loss of feedback control of cholesterol synthesis, a comparison was made of the properties of 3-hydroxy-3-methylglutaryl coenzyme A reductase after solubilization and partia...

Effect of neonatal modulation of cholesterol homeostasis on subsequent response to cholesterol challenge in adult guinea pig.

Experiments were designed to study whether or not the mechanism of handling dietary cholesterol in adulthood can be modulated by the manipulation of cholesterol homeostasis during neonatal period. The effects of enhancing cholesterol degradation (cholestyramine feeding), high dietary cholesterol intake, and early weaning during neonatal period of guinea pigs on their subsequent plasma cholester...
